Van Nuffel was influential in the development of 20th-century church music, particularly in Belgium. He was a member of the Caecilia-Concert, a group of Belgian composers who sought to promote sacred music. He also worked with Flor Peeters, another Belgian composer and organist, in preserving the tradition of Gregorian chant in church music.
Van Nuffel's compositions are characterized by their harmony, rhythm, and use of polyphony, and he is particularly known for his choral works. His most famous compositions include the Missa in Simplicitate, Missa in Honorem Sancti Dominici, and Missa Laudate Dominum.
